The world lies in ruin. Zombies and horribly mutated monsters roam free. All of this is the fault of the Umbrella Corporation, who still continue their unethical research from various labs beneath the streets across the globe. Alice (Milla Jovavich) continues her fight against them, and now picks up a search for her friends, who left for Alaska, but have not been heard of in six months. Upon venturing to Alaska, Alice finds a confused Claire Redfield (Ali Larter), who attacks her because of mind altering drug being injected into her. Removing the scarab on Claire's chest that has been injecting her with the drug, Alice discovers the effects of the drug also cause some type of memory loss. Flying to the "City of Angels", Hollywood, Alice and Claire take refuge in a prison, soon finding other survivors inside, one of which claims to be Chris Redifeld (Wentworth Miller), Claire's brother. While in the prison, Alice, Claire and the other survivors, discover that Arcadia, the place that promised safety in Alaska, is actually a tanker, roaming the ocean not far away from the prison. As a huge mutated Axeman busts his way into the prison, allowing the zombies to freely enter, the group is forced to travel through the sewers in an attempt to reach Arcadia. After many of their fellow survivors are killed, lost or already gone, Alice, Claire and Chris make their way to Arcadia, to find it is in fact another base for the Umbrella Corporation, luring people there with their false radio broadcasts for experimentation. On board, the group finds Umbrella chairman, Albert Wesker (Shawn Roberts), and while Alice is preoccupied, Claire and Chris must fight the super-fast and super-strong man. Upon his defeat, the group releases the people who would have been experimented on, including fellow Las Vegas survivor K-Mart (Spencer Locke). While the group watches over the released prisoners, Wesker makes his escape on a jet. Removing an explosive from the tanker and placing it on the jet, Alice, Claire and Chris watch Wesker's  plane explode. What they don't see, however, is a lone parachute flying away from the explosion. Very soon after, dozens of Umbrella soldiers come flying onto the tanker, to fight the group, led by Alice's lost comrade, Jill Valentine (Sienna Guillory), who is under the same control Claire was.
